Glutethimide and circulating 1,25-dihydroxyvitamin D in vitamin D intoxication.

چکیده
Iqbal and Taylor used glutethimide to treat a patient with vitamin D2 intoxication, reasoning that induction of hepatic microsomal enzymes by the drug would result in more rapid catabolism of vitamin D metabolites. The treatment corrected the hypercalcaemia, and the raised serum total 25-hydroxyvitamin D concentration fell with induction of the enzymes, as indicated by a rise in y-glutamyltransferase activity, but rose again when induction was stopped. Lukaszkiewicz et al successfully treated vitamin D3 intoxication in a 7 month old boy using phenobarbitone and phenytoin to induce hepatic microsomal enzymes.2 We report further analyses of the vitamin D metabolites in Iqbal and Taylor's patient.

Serum 1,25-dihydroxyvitamin D3 concentration and renal 25-hydroxyvitamin D 1 a-hydroxylase activity were measured in rats fed various levels of calcium, phosphorus and vitamin D3. Both calcium deprivation and phosphorus deprivation greatly increased circulating levels of 1,25-dihydroxyvitamin D3. The circulating level of 1,25-dihydroxyvitamin D3 in rats on a low-calcium diet increased with incr...
متن کاملVitamin D: its role in cancer prevention and treatment.
Vitamin D, the sunshine vitamin, has been recognized for almost 100 years as being essential for bone health. Vitamin D provides an adequate amount of calcium and phosphorus for the normal development and mineralization of a healthy skeleton. Vitamin D made in the skin or ingested in the diet, however, is biologically inactive and requires obligate hydroxylations first in the liver to 25-hydrox...

Background & Aims: 1,25-dihydroxyvitamin D3 (1,25 (OH)2 D3), the biologically active form of vitamin D, exerts an immunosuppressive effect through binding to its specific nuclear receptor. The present case-control study was done to examine the possible association of BsmI polymorphism in vitamin D receptor gene (VDR gene) with severity of multiple sclerosis (MS). Methods: 267 Iranian patients w...
متن کاملEffects of Vitamin D in Neonates and Young Infants
Vitamin D is important for the development, growth, and mineralization of the skeletal in neonates and children. Vitamin D is essential for intestinal absorption of calcium. Vitamin D is metabolized to 25-hydroxyvitamin D and then to 1,25-dihydroxyvitamin D. Pregnant women in developed countries often have insufficient serum concentrations of 25-hydroxyvitamin D and a supplementation of vitamin...

We describe a simplified assay for 24,25-and 1.25-dihydroxyvitamin D in human serum. It involves two preparative steps, and normal chick intestine is used in preparing cytosol-binding protein. Our results for 24,25-dihydroxyvitamin D include a reference interval of 2.9--16 nmol/L (1.2--6.7 microgram/L), a mean of 6.7 nmol/L (2.8 microgram/L), an intra-assay CV of 11%, and an interassay CV of 22...
